Extracellular calcium induces neurite outgrowth through calcium-sensing receptors in PC12 HS cells

PC12 cells have been used extensively to study neuronal differentiation and neurite outgrowth. However, the effects of extracellular Ca2+ on not yet sufficiently evaluated. In this study, we investigated effect calcium using HS cells, which high sensitivity nerve growth factor (NGF). The addition (1 mM) medium induced outgrowth increased intracellular concentration. contrast, calcium-sensing receptor antagonist NPS2143 suppressed concentration, suggesting that promotes through receptors. An increase in concentration phosphorylation Akt cyclic AMP response element-binding protein (CREB). Semi-quantitative RT-PCR showed elevated mRNA expression brain-derived neurotrophic (BDNF) platelet-derived (PDGF), are regulated by CREB activation. Our results suggest can promote receptors, resulting promotion neurites. a useful model investigate regulation via receptors cells.
